[Abstract] Objectines to detect different cytokines (TGF-β、MCP-1、TNF-α、ICAM-1、 ICAM-2) concentration after LPS stimulation airway epithelial cells at the same time, to explore the role of LPS in the airway epithelial cells secreting a variety of cytokines. Methods A549 cell strain was cultivated and interfered by different concentrations LPS after 2h、4h、8h、24h and 30h. We collected culture medium and total protein in the cell,detected volume of cytokines TGF-β、MCP-1、TNF-α、ICAM-1 and ICAM-2 secreted by A549 cell using enzyme-linked immunosorbent assays (ELISA),to analysed the changes of above-mentioned cytokines at the different time period. Results 1. LPS can interfered the airway epithelial cells secreting TGF-β,MCP-1,TNF-α,ICAM-1 and ICAM-2;the secretion of ICAM-1 peaked when the concentration of LPS was 20ug/ml and the time was 4h and 8h; the secretion of MCP-1 peaked when the concentration of LPS was 50ug/ml and the time was 4h. 2. The volume of cytokines TGF-β,MCP-1,TNF-α,ICAM-1,ICAM-2 was related to the concentration and the period of LPS, Compared with the control group, the maximum secretion of TNF-α、MCP-1 and ICAM-1 were increased at all the different time, the difference was statistically significant ( P < 0.001 ); whereas at the 30 hour, the secretion of TGF-β was decreased and the secretion of ICAM-2 was not different from the control group(p>0.05), Compared with the control group, the maximum secretion of TGF-β and ICAM-2 were increased at all the other different time, the difference was statistically significant ( P < 0.001 ). Conclusions 1. At the different time, the different concentration LPS influenced the volume of cytokines TGF-β、MCP-1、TNF-α、ICAM-1 and ICAM-2. 2. At all the different time, an optimal concentration of LPS increasead the secretion of inflammatory cytokines TNF-α, MCP-1 and ICAM-1 and enabled them reaches maximum in airway epithelial cells; Within 24h ( including 24h), an optimal concentration of LPS increasead the secretion of inflammatory cytokines TGF-β and ICAM-2 and enabled them reaches maximum in airway epithelial cells.
